The first incarnation of NY 401 followed present day Columbia CR 20. The original designation was removed in the early 1950s.

The more recent designation of NY 401 replaced NY 245 along first completed section of the Genesee Expressway...what is now I-390...between Wayland and Dansville. The Interstate designation replaced NY 401 in 1974.
